Gel Notes

Method Essay / Green Channel

The Bands Were the Argument

Why the quiet grammar of lanes, ladders, and faint smears still decides what a lab believes before lunch.

I spent two weeks last winter repeating a failed ligation that looked fine in the spreadsheet and guilty on the gel. Lane four carried a bright shoulder where the insert should have been clean; lane five had the soft smear of a tired enzyme. The machine did not explain itself, but it refused to flatter us.

A readout with no patience for narrative

Every lab has a ritual for naming the bands: ladder, digest, control, maybe the sample nobody trusts. By the time the image exports, the decision has usually happened at the bench, in the green light, with a gloved finger moving from well to well and stopping where the story breaks.
